Bioflocculants and Their Applications

Bioflocculants are biodegradable polymeric materials that are generated during the development stages of a variety of microorganisms, such as fungus, actinomycetes, and bacteria. These materials have unique benefits such as biodegradability, efficient flocculation, and safety for the environment, which makes them a viable substitute for a range of industrial uses. Their non-toxic properties to people and the environment only serve to increase their allure

In a recent study, researchers isolated 39 bacterial strains from wastewater samples collected from Oyo and Osun states. These isolates were tested for their bioflocculant production abilities using a kaolin solution (5 g/L) as the test medium. The results indicated significant variations in the flocculating efficiency of the isolates, 

with the percentage of flocculating activity ranging from 3.00% to 75.63%. Among these, isolate TB3 exhibited the highest flocculating activity, followed by TM3, AM6, and API, while isolate AP3 demonstrated the lowest activity. Six isolates with the most effective flocculating properties were identified phenotypically as Enterobacter AM6, Streptococcus AP1, and others.

Materials and methods

Collection of Papaya Leaves

The College of Basic and Applied Sciences at Mountain Top University in Ogun State provided fresh Carica papaya leaves weighing 335.80 g. The leaves were properly cleansed with distilled water and dried prior to usage.

Extraction of Crude Enzyme

Five separate batches of 50.0 g of papaya leaves and 100 mL of ice-cold 0.1 N phosphate buffer (pH 5.5) were combined to extract the enzyme. After mixing and sifting the homogenate, the remainder was disposed of.The 500 mL filtrate was centrifuged for 10 minutes at 4 °C at 3000 rpm. One milliliter of cold phosphate buffer (pH 5.5) was used to wash the sediment, and the crude extract was then kept at 4°C for ten minutes.

Formation of Ammonium Sulphate Suction

The crude extract was saturated to 70%, then ammonium sulfate was added, and it was chilled for the entire night at 4°C. The mixture was separated from the supernatant the next day by centrifuging it for five minutes at 3000 rpm and 4°C.

One kind of globular cysteine protease enzyme called papain is well-known for having a single polypeptide chain structure that contains a sulfhydryl group that is essential to its proteolytic activity as well as three disulphide bridges. Because of its many uses, this enzyme is important to both the medical field and the food business.

This study set out to extract, purify, and analyze the papain found in Carica papaya leaves. At Nigeria’s Mountain Top University, papaya leaves were used to make crude enzyme extracts. 

The purification process involved a three-step method, utilizing 70% ammonium sulfate ((NH4)2SO4) and a DEAE-Cellulose column to achieve a high level of enzyme purity.

Recent developments in synthetic biology and enzyme discovery have greatly improved metabolic engineering, opening up a wider spectrum of substrates for use. The current developments with the industrial yeast Yarrowia  are highlighted in this review. This yeast is well-known for its versatility in using carbon sources, and it has demonstrated potential in the production of chemicals and biofuels. As such, it is an important component of the continuous attempts to improve biotechnological processes.

Even when bioconversion yields are high, many biotechnology processes struggle with economic viability due to the fluctuating and often high costs of commonly used substrates. To counter this, raw and inexpensive carbon sources, typically complex sugar polymers, are preferred. These substrates can be pretreated with chemical or enzymatic methods to break them down into simpler subunits, but more efficient, consolidated bioprocesses that streamline these steps are increasingly sought after.

Unfortunately, most industrial microorganisms are not naturally equipped to degrade raw substrates like lignocellulosic biomass or certain monosaccharide subunits. To address this, metabolic engineering has made significant strides, allowing scientists to modify these organisms so they can fully break down these complex substrates.
